Evaluation of Selected Gene Expressions of the Matrix Metalloproteinases and Their Inhibitors in Lens Epithelial Cells in Patients with PEX Syndrome

Summary: Purpose: To compare gene expressions of the selected matrix metalloproteinases (MMPs) and their tissue inhibitors (TIMPs) in lens epithelial cells in patients with PEX syndrome qualified for cataract surgery.
Material and Methods: 51 patients, 17 – with PEX (mean age: 78.9 ± 8.37) and 34 – without PEX (mean age: 72.4 ± 10.02), were included in the study. The anterior lens capsule specimens were obtained during cataract surgery. Levels of MMP-1,2,7,9 and TIMP-1,2 mRNA were measured using real time-PCR (real time-polymerase chain reaction) kits.
Results: In both groups MMPs genes activity in lens epithelial cells were at low, whereas TIMPs – at high levels. The significantly higher activity of MMP-7 was found in the PEX group (p = 0.03). Gender and intraocular pressure values had no influence on MMPs and TIMPs expression. The activity of MMP-9 decreased with age (p = 0.05, R = -0.27).
Conclusions: The significantly higher level of MMP-7 activity and probably changes in the MMPs/ TIMPs balance in LEC may promote the development of PEX syndrome.
