Effect of Ketotifen Ophthalmic Solution on Eosinophil Cationic Protein Concentration in Tears of Patients with Allergic Conjunctivitis

Summary: Aim: Eosinophil cationic protein indicates the allergic reaction intensity, as well as in serum and tears. The aim of this study was to estimate alteration in ECP concentration in tears, before and after treatment with ketotifen ophthalmic solution.
Methods: tears were collected from 18 patients with seasonal allergic conjunctivitis or perennial allergic conjunctivitis. The investigation of the patients included history of atopy. None of the patients received any systematical or topical medicines. Clinical signs of ocular allergy were evaluated using Abelson scale. Shirmer’s tests, tear break-up time and skin prick tests also were carried out. Tears were collected from the lateral canthus of one eye in each subject in 50 µl before and after 20 days of treatment. The samples were kept at –20°C until they had been analyzed. Eosinophil cationic protein amount in tears was analyzed with enzyme–immunoassay for ECP (Pharmacia CAP 100E System UniCAP ECP).
Results: patient’s ocular allergy clinical symptoms intensity correlated with ECP tear concentration alteration. After treatment, signs of allergic conjunctival reaction subsided. We noticed increased congestion intensity, itching and tearing only in one patient. Four of the patients did not show up for the follow–up visit after treatment.
Conclusions: all patients with allergic conjunctivitis (except one subject) showed reduced symptoms of allergic reaction after treatment with ketotifen ophthalmic solution. Symptoms and the tear ECP amount correlated significantly in all examined patients. Due to the small number of samples (high costs), those studies required further investigation.
