Summary: Fluorescein has been widely used as a diagnostic dye in ophthalmology and optometry for over a century. It enables corneal and conjunctival abnormalities visualization, retinal angiography and Goldmann applanation. Fluorescein has fluorescent properties. The intensity and colour of ocular surface fluorescence varies according to fluorescein concentration, the thickness of the fluorescien layer, the change of pH and the wavelength of the exciting light form the illumination light source. There have been many attemps to explain the mechanisms of ocular surface fluorescein staining. The main theories are: surface pooling, uptake by the cells and ingress around the cells. It is proved that fluorescein is absorbed mostly by living epithelial cells. Its highest concentration is found in cells that are at the stage of apoptosis. In dead epithelial cells the concentration is the smallest. Since the phenomenon of fluorescein ocular surface staining is still not fully understood further research is necessary.
